It's because we are Micro Biologists now Nicki…..its how we do. But also, it's to eliminate any doubt pertaining to the growth of E. Coli on the plates containing the T4 bacteriophages. If you were to do 5-10 tubes and nothing grew that's fine, but one could argue that there is still a small chance that E. coli could grow on the T4 bacteriophage plates because our tests weren't extensive enough. With 100 tubes, were eliminating that argument and further supporting our hypothesis.
But mostly its because we're Micro Biologists :)
